Element AVITI System

Revolutionizing Benchtop NGS Systems.

From Sequencing by Synthesis (SBS) to Avidity Base Chemistry (ABC).
Next-Generation Sequencing (NGS) has transformed our understanding of biology, impacting human health, as well as the health of animals, plants, and microorganisms. While past advancements have ushered in a brilliant era of genomics, the core technology has since stagnated. Enter Avidity Base Chemistry (ABC) – an innovative sequencing technology that leverages the power of avidites to surpass and enhance established models in terms of cost, quality, and flexibility.

AVITI™, the innovative sequencing system developed by Element Biosciences, based on ABC sequencing chemistry, utilizes Rolling Circle Amplification (RCA).
The fragment to be sequenced, initially linear, circularizes on the flow cell. Thousands of copies are then generated from this template, each originating from the original fragment, not as copies of copies (clonal copies). This innovation ensures that if an error is introduced during PCR, it is purely random and does not propagate across all copies, drastically improving data quality.

Q-Score of 40 or Higher (Q40+)
AVITI™ delivers high accuracy with a Q-score of 40 or higher, corresponding to an error rate of less than 1 in 10,000 bases. This ensures high-quality and precise sequencing, making the system ideal for applications requiring highly reliable data.

Dual Flow Cell: Two Sequencers in One.
At the heart of the sequencer is the dual independent flow cell, a feature unique among benchtop sequencers. Thanks to the dual independent flow cell, AVITI™ ensures high operational flexibility and the ability to double data throughput, increasing overall performance.

Dual Independent Lane per Flow Cell: Even More Flexibility.
The flexibility of the AVITIis further enhanced by the independent loading capability of the two lanes within each flow cell. This allows different experiments to be loaded and run independently and in parallel.

  • Optional Upgrade for separate loading of libraries/pools in each lane within a single flow cell
  • Increased Multiplexing Options
  • Greater Flexibility for sample volume throughput variability
  • Improved Control over read distribution within pools
  • Available for both AVITI and AVITI LT systems

Flow Cells Working with Cloudbreak Freestyle™ Chemistry.
Three different flow cell configurations employing Cloudbreak Freestyle™ chemistry are available to meet various needs:

  • Low Output: Ideal for small projects and laboratories with few samples, allowing in-house sequencing and quick runs.
  • Medium Output: Balances throughput and cost for intermediate-sized projects and labs.
  • Large Output: Perfect for large projects and labs with many samples, minimizing cost per sample.

Incredible Scalability: AVITI™ Can Grow Anytime.
AVITI™ is available in both full” and LT (Low Throughput) versions, but the instrument remains the same. Acquiring it represents a long-term investment. At any time, the base version can be upgraded to the more advanced version, keeping costs contained and avoiding cumbersome equipment changes.

Competitive Pricing and Top Performance.
Element Biosciences, with its new ABC sequencing chemistry, has completely changed the NGS landscape. Despite outperforming sequencers in the same category, it has a lower cost per sample, comparable to the most productive instruments on the market that require multi-million euro initial investments. With this disruptive policy, Element enables laboratories of all sizes and budgets to do more science.

Universal Compatibility.
Cloudbreak Freestyle™, the sequencing chemistry used by AVITI™, is fully compatible with all currently available libraries, requiring no conversion. This ensures laboratories do not need to change their operational routines or make additional purchases.

  • Operational Flexibility: It allows for greater operational flexibility, facilitating use in various laboratory settings.

The AVITI system minimizes error and ensures a Q-score of 40 or higher thanks to several key factors that enhance sequencing accuracy:

  • Advanced Sequencing Technology: AVITI™ uses Avidity Base Chemistry (ABC) technology, which employs avidins for base detection. This methodology enables stable and reliable reading of DNA bases during the sequencing process.
  • RCA and Polonies: The use of Rolling Circle Amplification (RCA) helps maintain the integrity of the original template, reducing the propagation of errors during amplification and polony generation.
  • Separation of Detection and Advancement Phases: AVITI™ clearly separates the base detection and template advancement phases. This approach optimizes enzymatic conditions for each phase, minimizing the possibility of errors during nucleotide incorporation.
  • Minimization of Sequencing Errors:
    • Low Reagent Concentration: It uses nanomolar concentrations of reagents, which not only reduces costs but also lowers the likelihood of errors during nucleotide incorporation and improves detection specificity.
  • High Data Quality:
    • Elimination of PCR Errors: Unlike cluster generation in traditional SBS, AVITI uses RCA, avoiding error propagation associated with PCR and enhancing sequencing fidelity.

User Benefits: These combined elements allow AVITI to sequence with a Q-score of 40 or higher, resulting in an extremely low error rate (less than 1 error per 10,000 bases sequenced). This precision is essential for critical applications such as precision medicine, genetic research, and other areas where data reliability is crucial for clinical and scientific decisions.
